BIOL1020 restriction enzyme digest simulator
In this simulator you will see the sequence of the part of the Tas2r38 taste receptor gene that you amplified in a previous session. You will see both a taster variant and a non taster variant, so this is what would be amplified from someone who is a heterozygote for this trait.
To simulate adding a restriction enzyme, click the 'RE' button in the bottom left and input the enzyme recognition site, with an asterisk to represent the backbone cut location. This is standard terminology used for all restriction enzymes. Therefore, to simulate the HAEIII enzyme we used, type GG*CC into the RE box. Then drag the enzyme to the DNA and see what happens!
We have also given you the primer sequences we used, so you can remind yourself how PCR was used to obtain millions of copies of these sequences. Just remember that we didn't add the restriction enzyme until AFTER the PCR was finished.
